Abstract

Flavonoid quercetin is capable of scavenging free radicals and protecting cells from oxidative injuries. It may provide therapeutic potential for the prevention and treatment of neuronal disorders caused by oxidative stress. In this study we used P19 neurons, originated from the mouse embryonal carcinoma P19 cell line, to investigate the effects of quercetin on H2O2-induced neuronal death. During the oxidative injury, quercetin increased the number of viable cells. Quercetin decreased leakage of lactate dehydrogenase from cells exposed to 1,5 mM H202, suggesting protective role of quercetin on necrosis. Activation of caspase-3/7 induced by 150 μM H202 was inhibited in the presence of quercetin, thereby demostrating its antapoptotic activity. However, it neither affected 150 μM H202- induced decrease in expression of Bcl-2 mRNA, nor the expression of Bax mRNA that was not affected by 150 μM H202. In conclusion, our results indicate neuroprotective potential of quercetin against oxidative stress-induced neuronal cell death.
