Abstract

Family Hydrophilidae is large family of water beetles with morphologically diverse representatives that live in various habitats. According to the fact that hydrophilida are poorly investigated in Croatia, field research was carried out in different areas of country in the period from May 29th, 2010-August 25th, 2011. The main objective of this study was to examine and clarify evolutionary relationships within the family Hydrophilidae, expecting that it would help in revisions this family is currently undergoing. Samples were determined using morphological characteristics. DNA was isolated from the samples and phylogenetic analyses were conducted by the use of two mitochondrial markers: genes for cytochrome oxidase subunit 1 (COI) and for smaller subunit of ribosomal RNA (12S rRNA). The results of research indicate that COI and 12S rRNA provide good distinguishing features on lower levels (species and genus) and that they are suitable for analyses on that level, but not sufficiently informative for analyses of deeper phylogenetic reltionships (on the subfamily and family level). Additionally, it has been found that analyses are more reliable if they are conducted with joined molecular markers (COI and 12S rRNA). This research provides the basis for the future studies, both taxonomic and phylogenetic, on hydrophilids in Croatia and Europe.
