Abstract

Plasminogen activation system is a proteolytic system that plays an important role in physiological and pathological processes in which extracellular matrix remodelling occurs. Plasminogen activation system is regulated on the level of transcription of urokinase-type and tissue-type plasminogen activator (uPA and tPA), plasminogen activator inhibitors (PAI-1 i PAI-2) and uPA receptor (uPAR). The aim of this study was to investigate the regulation of urokinase activation system in A1235 human glioblastoma cells after alkylation treatment and to determine whether this induction of urokinase activity was associated with activation of JNK MAP-kinase. A1235 cells, lacking the expression of the O6metilguanin methyltransferase, after alkylation damage induced increase in urokinase activity. Inhibitor JNK didn´t show significant inhibition of uPA activity in A1235 cells after alkylation damage. The experiments indicate the existance of complex pathways regulating urokinase activity, showing the cell type specificity.
