Abstract

Osteoporosis is a disease characterised by the loss of bone density and deterioration of bone micro architecture, leading to an increased risk of fracture. The aetiology of osteoporosis is varied whereby postmenopausal osteoporosis is the most common form. Several classes of drugs are available for the treatment of osteoporosis: antiresorptive drugs, oestrogen, selective oestrogen receptor modulators, bisphosphonates, parathyroid hormone, and strontium ranelate. Due to the adverse effects of these drugs, a natural alternative offered in the form of phytoestrogens is of interest. The present study was designed to investigate the possible protective effect of preparations of chysin against retinoic acid -induced osteoporosis in rats. Oral administration of chrysin at doses of 100 mg kg-1 and retinoic acid at dose of 80 mg kg-1 to rats for a period of 15 days resulted in a significant increase the content of bone ash, calcium, and phosphorus content in rats; chrysin prevented osteoporosis induced by retinoic acid. In addition, blood samples were collected after treatment of rats with test components to analyze DNA damage and number of micronucleus in reticulocytes of peripheral blood by standard comet assays and micronucleus test. The results from the genotoxicity studies suggest chrysin is safe for consumption with no observed adverse effect on peripheral lymphocytes. These outcomes suggest that chrysin offer a potential in the management of osteoporosis in vivo.
