Abstract

In vitro assays of pesticides are the fastest and easiest way to determine the toxic potential of certain pesticides. The aim was to determine how the pesticides imazalil, cypermethrine and carbendazime and their combinations influence erythrocyte hemolysis in vitro compared with hematotoxic potential in vivo in Swiss-mouse. Hemolytic potential in a in vitro experiment was measured with the modified method according to Grinberg. Erythrocytes obtained from mouse tail veins were washed and resuspended (2% v / v) in isotonic solution of PBS with 5.5 mm glucose. In each solution pesticides and their combination were added from 5 μM to 10 mM. The percentage of hemolysis was determined spectrophotometrically. In in vivo experiment animals were exposed to the NOAEL doses of the each individual pesticide and their combinations (NOAEL, 1:1). Pesticides were applied orally during 28 consecutive days. Following hematological parameters were determined: the red blood cell count RBC, HCT hematocrit, hemoglobin HB. Imazalil and its combinations showed the strongest hemolytic potential in in vitro experiment at all concentrations. In in vivo experiment, after 28 days treatment with imazalil and cypermethrine caused polycythemia. Carbendazime did not significantly change the number of red blood cells.
