Abstract

Increasing amount of toxic and mutagenic substances in the environment is caused by rapid industry development. The most obvious pollutants are heavy metals like copper, zinc and cadmium. Since heavy metals can be accumulated by plants, they become available to other organisms through food chain including animals and men. The main goal of this research was to determine genotoxicity of copper, zinc and cadmium individually and in mixture by comet assay as a non-specific genotoxicity biomarker in tobacco, Nicotiana tabacum L. Bioaccumulations of heavy metals was measured with inductively coupled plasma atomic emission spectroscopy (ICP-OES). Comet assay sensitivity was verified with treating tobacco cell nuclei in in vitro conditions with hydrogen peroxide as a model genotoxicant. Tobacco seedlings were exposed to different concentrations and combinations of heavy metals for one month. Results showed mild indirect genotoxicity of investigated heavy metals. Tobacco cell nuclei were exposed in vitro to different concentrations and combinations of investigated metals for two hours. Results showed mild direct genotoxicity. The results implied influence of cadmium to bioaccumulation and genotoxicity of copper and zinc. The mechanism of their interrelation could not be concluded because intake mechanism is very complex therefore further investigations are needed.
