Mesenchymal stem cells (MSCs) are nonhematopoietic multipotent stem cells mainly residin in the bone marrow (BM). MSCs show immunomodulatory properties in vitro that make them intersting in the field of cellular therapy. MSCs have ben clinically applied to improve engraftment in hematopoietic stem cell transplantation and to treat acute graft versus host disease. Incidence of MSCs in BM is very low and ex vivo expansion of MSCs is necessary. We expanded MSCs in vitro in media supplemented with 10% human plateet lysate (PL) thus avoiding standard use of animal origin supplements. MSCs isolated from BM samples of healthy donors were sucessfully cultured in medium containing PL. During propagation period of 35 days, MSCs continuously expressed mesenchymal characteristics: they adhered to plastic surface shoing typical firoblastod shape, they expressed mesenchymal immunophenotype and formed fibroblast colonies. These findings give basis for further efforts in evaluating use of PL i MSCs expansion methods.
