Abstract

Development of different cell types from the zygote of multicellular oraganism is based on the establishment of the the cell polarity and asymmetric cell division. Protein TaMAB2 was found in the zygote and early embryonic stages of wheat. In this theisis influence of the protein domains of TaMAB2, MATH and BTB, on it's charactristic polar localisation on nuclear envelope was analysed. For this purpose GFP tagging of proteins was used for plant cell transformation. Deletions were made by using PCR. It was found that deletion of each domain influence the protein localisation by disturbing its polar distribution. While deletion of the MATH domain disturbed polar distribution in all BMS cells, deletion of the BTB domain changed polar localisation of TaMAB2 in 74% of BMS cells. Analysis of epidermal onion cells showed similar results. Precise role of TaMAB2 in establishment of the plant cell polarity and asymmetric cell division is still unknown.
