Insufficient effectiveness of current therapy of cancer patients has stimulated the search for new, more effective tretament, including the screening of new compounds. To increasae the beneficial characteristics of diazencarboxamides, new compounds, diazendicarboxamides have been synthesised. The theme of this graduation thesis was to investigate their biological activity. The most effective among them was JV-158, as determined by MTT assay on cervical carcinoma HeLa cells. Its cytotoxicity was tested further on several tumor cells lines showing that Hela cells were the most sensitive, while colon carcinoma HCT-116 cells were most resistant. Depletion of intracellular glutathione level did not change the sensitivity of HeLa cells to JV-158, indicating that GSH was not involved in cell response to this compound. Regarding the type of induced cell death, JV-158 triggers necrosis. Based on these results we can assume that the compound JV-158 could be a new potential anti-cancer drug.
