Abstract

Several molecular methods are applied for the classification of bacteria. These methods are based on nucleic acid hybridization of an unknown bacterial strain of interest with that of a closely related already identified strain to determine the degree of genetic complementarity. The degree of hybridization, proportional to the degree of similarity is used to establish taxonomic identification. The broadly used techniques in bacterial taxonomy are DNA-DNA hybridization, DNA-rRNA hybridization, oligonucleotide hybridization and DNA fingerprinting. Here are described some of the most fundamental techniques used in bacterial classification with an emphasis on methodological issues and differences between experimental approaches.
