Oxidative stress is one of the key factors in the development and progression of many neurodegenerative diseases. The aim of the present study was to explore the effects of the flavonoid quercetin, one of the most potent antioxidants, on the survival of P19 neurons under conditions of oxidative stress provoked by excess copper ions. In moderately injured neurons exposed to 0.5 mM CuSO4 for 24 hours, 150 μM quercetin increased viability and reduced copper-induced increase in the production of reactive oxygen species (ROS) and the caspase-3/7 activity. Quercetin also decreased copper-induced depletion of glutathione content. Effect of quercetin on survival of P19 neurons was prevented in the presence of wortmannin, an inhibitor of the PI3K/Akt signaling pathway. Treatment with 1 mM CuSO4 induced further decrease in neuronal survival, together with very pronounced ROS accumulation and activation of caspase-3/7, as well as decreased expression of antiapoptotic gene Bcl-2 and increased expression of proapototic gene Bax at the transcriptional level, but the toxic effect of copper was not reduced in the presence of 150 μM quercetin. The obtained results suggest that neuroprotective effect of quercetin against moderate oxidative injury induced by excess copper ions are related to its antioxidative and antiapoptotic activity, as well as modulation of the PI3K/Akt signaling pathway.
