Abstract

Cardiovascular diseases (CVDs) are the major cause of morbidity and mortality worldwide, and the susceptibility to developing CVD is increased by the interaction of several environmental, biochemical and genetic risk factors. Since longevity is nothing but avoiding death, the association analysis of polymorphisms in four candidate genes for CVDs (ACE, APOE, eNOS, MTHFR) with non-genetic risk factors was performed in a sample od people aged 80 years and over. Logistic regression analysis showed that the combination of ACE-DD and MTHFR-CC genotypes reduced the chance of survival into nonagenarian age, in a model explaining as much as 27.5% of the variance. The meta-analysis, carried out using published data for the European populations, tested the frequency of the investigated alleles between general and 80+ population, and showed that ACE D allele and APOE E2 allele were more common in elderlies, while E4 allele was more common in general population. This research indicated the existence of selective mortality in APOE E4-carriers, as well as the age- dependent, pleiotropic effects of the ACE I/D polymorphism.
