Abstract

The aim of this study is to evaluate the expression pattern at histological level of the chondroiotin-sulphate proteoglycan neurocan (NCAN) in the human fetal neostriatum from gestation weeks (GW) 12 to 40, using indirect immunohystochemical staining on postmortal brain tissue. Neurocan is expressed in the neostriatum from 16 – 25 GW. In the early phase of expression, from 16 – 18 GW, neurocan is expressed in the cell islands – striosomes, and at 19/20 GW shows a shift in expression from the striosomes to the surrounding matrix. After the 23rd GW neurocan expression declines untill GW 27 when its expression ceases. Neurocan shows the highest rate of expression between the 19th - 23 GW in a tight cell-free zone that surrounds the striosomes – the peristriosomes or perimeters, which are most pronounced during fetal and early postnatal development. We propose that the selective expression of neurocan in the different modules of the fetal striatum reflects the differences in the sequence of growth and maturation of striatal afferents, and/or that neurocan has a role in forming the boundaries between striatal modules.
