Abstract

Diffuse large B-cell lymphoma (DLBCL) is a neoplasm of B-cells with a characteristic diffuse pattern of distribution. It accounts for approximately 30% of non-Hodgkin lymphomas in elderly. DLBCL can be stratified, based on gene expression profiling, into GCB (germinal centre B-cell like) and non-GCB (non-germinal centre B-cell like) subtypes. In most DLBCL cases, a large number of genetic rearrangements can be detected. The most frequent is BCL6 translocation, followed by translocations of BCL2 and c-MYC. The aim of this study was to analyse changes in the amount of proteins involved in the replication process and the chromatin reassembly mechanisms in DLBCL subtypes. These changes might be responsible for aberrations in replication accuracy which further leads to accumulation of additional mutations in tumor cells causing tumor progression. DLBCL cases were classified into GCB and non-GCB subtyes using immunohistochemical algorithm by Hans et al. RNA from all samples was extracted and converted to cDNA by reverse transcription. q-RT PCR analysis of DNMT1, PCNA, MCM2, MCM7, GMNN, CDT1, EZH2 and p300 expression showed an increased expression of CDT1 and DNMT1 as well as a decreased expression of PCNA, MCM2, MCM7 and EZH2 in GCB when compared with a non-GCB subtype. Results suggest contribution of replication and chromatin reassembly machinery components to DLBCL pathogenesis.
