Abstract

Follicular lymphoma is a B–cell neoplasm composed of cells originating from follicular centres (germinal centres) of lymph node that express BCL6, CD10 and, usually, BCL2. The translocation t(14;18)(q32;q21), which leads to enhanced expression of antiapoptotic BCL2 protein, is the characteristic type of genetic alteration in follicular lymphoma. Follicular lymphoma is the only lymphoma that has grades - stages of progression. One of the potential developmental pathways of higher stages of progression ( there are 3 stages) might include the process of replication and the reassembly of chromatin structure after replication. Aim of this research was to analyze the transcript levels of genes which are important for the process of replication and the re–establishment of chromatin in tumor and non–tumor part of tissue affected with the neoplasm. Ten samples of lymph nodes with follicular lymphoma grade 2 and 3 were used. Tumor and non– tumor part of the tissue were selected and gathered using laser–microdissection technology, RNA was isolated and reversely transcribed into cDNA for QPCR analysis of DNMT1, PCNA, MCM2, MCM7, CDT1, EZH2, GMNN and p300 transcripts. The results showed no significant differences in the expression of the analyzed genes between groups of tumor and, surrounding, non–tumor tissue of affected lymph nodes.
