Abstract

Earthworm Eisenia fetida was exposed to increased concentrations of silver in nanoparticle and ionic (AgNO3) form. The comparison of bioaccumulation between two forms of silver, along with comparison of similarities and differences in subcellular and cytosolic distribution of silver after exposure were made. The potential of both silver forms to induce the synthesis of metallothioneins and change the distribution of chosen essential elements (Cu, Zn, Mn, Fe, Co, Mo, Se) and Cd between the particulate (pellet) and cytosolic fraction of E. fetida tissue was investigated. The distribution of chosen elements among different molecular mass cytosolic fractions separated by size exclusion HPLC was also analysed. It was shown that both silver forms accumulate in earthworm tissues. Bioaccumulation levels were higher in earthworms exposed to ionic silver when compared to nanoparticle silver. There are significant differences in cellular and cytosolic distribution of silver between these two forms of exposure. Silver nanoparticles are not inducing metallothionein synthesis while ionic silver induces it, but only in the highest concentration of exposure. Part of intracellular silver is bound to metallothioneins which are already expressed in cells. The quantitative and qualitative differences in intracellular distribution of essential elements iron, cobalt and molybdenum show potential as biomarkers of nanoparticle and ionic silver form exposure.
