Abstract

Pytopathogenic fungus Cryphonectria parasitica causes blight of chestnut (Castanea sativa). In Europe the disease is controlled by the presence of virus Cryphonectria hypovirus 1 (CHV-1), which reduces fungal virulence and sporulation, causing the phenomenon called hypovirulence. In this research three CHV-1 isolates belonging to the two subtypes of the virus were transferred to virulent fungal strains. The presence of virus was confirmed by dsRNA isolation, reverse transcription and amplification of two viral cDNA open reading frames (ORF-A and ORF-B). The activity of intracellular laccase LAC-2 and extracellular laccase LAC-1 in virulent and hypovirulent isolates of the fungus was compared. The results have shown that the virus CHV-1 does not affect the activity of the intracellular laccase LAC-2, whereas the activity of the extracellular laccase LAC-1 depends on the subtype and isolate of the virus and genotype of the fungus.
