Abstract

Testicular cancer (TC) represents about 1% of all new diagnosed malignant cancer in male population. TC are believed to arise from primordial germ cells in early embrional development when DNA methylation and parental imprints are erased and totipotency is restored. With differentiation of subgroups of TC the level of DNA methylation gradually increases. The main goal of this experiment was to analyse methylation level inside CpG islands of the MGMT and RASSF1A genes in blood samples of patients with testicular cancer and healthy individuals. For this purpose, methylation status was determined for each CpG sites of bisulfite converted DNA using pyrosequencing method. The results are not showing statistically significant change in CpG methylation within the analysed fragments of MGMT gene in patients with testicular cancer compared to healthy individuals. However, for RASSF1A the results show statistically significant change in CpG methylation analysed in patients with testicular cancer compared with healthy individuals, thus revealing the potential of this gene as a methylation biomarker in periferal blood.
