Abstract

Strawberry tree is a plant species which due to the rich polyphenol content finds its use in fitotherapy. The most represented phenol compound within the leaf of strawberry tree is arbutin. Natural products containing arbutin are most frequently used in urinoinfection therapy. Therapeutic effects of those products are based on antibacterial activity of hydroquinone, pharmacologically active metabolite of arbutin. This research was carried out with the aim of evaluation of genotoxic effect of strawberry tree leaf extract and arbutin on animal model. Rats of the Lewis breed were separated by gender and arranged into 6 groups by 4 animals (1 control and 2 treated groups for each gender). The test groups were treated with strawberry tree leaf extract or arbutin in concentration of 200 mg/kg during 14 days. For evaluation of genotoxic effects, we applied the standard performance of comet assay in alkali conditions and micronucleus assay in vivo. The comet assay was carried out on the cells of peripheral blood, kidneys and liver while micronucleus assay was carried out on the reticulocytes of peripheral blood. Measured levels of damage using comet assay do not exceed 10% DNA in comet tail for all of the cell types. Furthermore, we didn’t notice statistically relevant increase of micronuclei number as a consequence of applied treatments. Based on the indicated results we can conclude that the strawberry tree leaf extract and arbutin have low genotoxic potential.
