Abstract

Internal validation of a new method is mandatory step before its implementation into the everyday casework. AmpFlSTR® SGM Plus® Amplification Kit is used for the analysis of traces and reference samples for forensic purposes. The aim of this study was to validate AmpFlSTR® SGM Plus® Amplification Kit at low DNA concentrations as follows: 0.06 ng/μL, 0.04 ng/μL, 0.02 ng/μL and 0.01 ng/μL from AmpFlSTR® DNA positive control, reference buccal swab sample and forensic epithelial cells sample. Full DNA profile was detected in positive DNA control in all dilutions. Also, the same was observed for reference sample except in concentration 0.01 ng/μL. Full DNA profile was found only at concentration 0.06 ng/μL in forensic sample. In addition, a partial DNA profiles were observed in all other concentrations tested.
